The Santo Domingo Church was rebuilt 5 times in Intramuros before finding its permanent home in Quezon, City. The first Santo Domingo church was built in 1588, collapsed in 1589 due to poor foundations. It was rebuilt in 1592, now in stone but burned in 1643. Constructed again but was caught between the massive quake of 1645. The 1692 church again succumbed to an earthquake in 1863. The fifth church was unveiled in 1868, designed by the first Filipino architect Felix Roxas Sr. in the neo-gothic style.When it was destroyed in 1941, the church relocated to its present site in Quezon, City. The former site in Intramuros was acquired by the government and part of it is now The Bank of Philippine Islands (BPI).

Intendencia or La Aduana was first constructed in 1823 and completed in 1829. The Spanish building was meant to house different government offices as well as attract merchants into entering Intramuros. The building was damaged in the 1863 earthquake which led to its demolition in 1872. After its reconstruction in 1876, it became the first Office of the Archives. The La Aduana also became the first home of the Philippine Senate. The structure suffered damages from World War II but after its reconstruction after the Manila Liberation, it housed the National Treasury, Central Bank of the Philippines and the Commission on Elections. It was around this time when it was renamed to Intendencia.In 1979, the Intendencia suffered fire and thus vacated. The facade of the building was restored in 1998 by the National Archives. More plans on its reconstruction are being discussed.

Originally called Plaza Aduana, the Plaza España is a little triangle open space made by the intersection of Andres Soriano Avenue, Solana Street, and Muralla Street. It was then renamed as Plaza de Los Martinez de la Integridad de la Patria or simply  Plaza de Martires in 1897, in honor of the Spanish soldiers that died in the Philippine Revolution. When the Americans came, they renamed it to Plaza España. The Intramuros Administration began restoring the plaza in 1982. A monument of King Philip II is erected at the center of the plaza in 1998. The plaza was inaugurated in 2000 as part of the Philippine Centennial.

Founded in 1611, the University of Santo Tomas is now the oldest university in Asia and the largest catholic university in the world on one campus in terms of population.Bishop Miguel de Benavides, the third archbishop of Manila founded the university using one thousand five hundred pesos and his personal library to build a college to prepare young men for the priesthood.During World War II, the university was converted to campus into an internment camp where around 2,500 allied civilians were detained. Other parts of the university were converted into living quarters. The university left Intramuros in 1927 to transfer to its current location in Espana Boulevard when the Dominicans thought their campus in Intramuros was getting smaller compared to their growing population.The former site of UST is now occupied by Banco Filipino Condominium.
